In search of loops in the cell nucleus (Physique Théorique) Understanding the functional architecture of the genome and its dynamics is one of the great challenges of systems biology. The three-dimensional arrangement in the cell nucleus of the protein-DNA assembly that makes up chromosomes appears to be crucial for the regulation of gene expression, and loop formation has recently emerged as a key regulatory mechanism. Rapid advances in experimental techniques now make it possible to directly explore these complex structures by imaging the spatial arrangement of sequential sites. From a modeling point of view, while polymer physics is a fundamental modeling ground, increasingly sophisticated analysis tools are becoming necessary to explore the data efficiently. I will present original approaches based on spectral analysis that have enabled us to better describe the organization of polymers, and to develop a method for identifying loops in chromatin.
