Microvessel blood flow imaging techniques are widely used in biomedical research and clinical diagnostics where many diseases have a vascular etiology or involvement. For testing purposes, zebrafish embryo provides an ideal animal model to achieve high-resolution imaging of superficial and deeply localized vessels. Moreover, the study of the formation of a closed circulatory system in vertebrates is a topic of recent interest in biophysics. However, most of the existing techniques are invasive due to the use of a contrast agent for imaging purposes. Recent developments in Digital Holography and Laser Doppler Holography techniques can be considered to alleviate this issue. Laser Doppler holography and transmission microscopy can be coupled to analyze blood flow in fish embryos by adapting a laser Doppler holographic setup to a standard bio-microscope: the two beams of the holographic interferometer (illumination of the object and reference), whose frequency offset is controlled, were addressed to the microscope by optical fibers. Multimodal acquisition and analysis of the data is made by acting on the frequency offset of the two beams, and on the location of the Fourier space filtered zone. In this work, we show that it is possible to select the signal of moving scatterers, and to image Red Blood Cells (RBCs) and blood vessels. We investigate the nucleation and propagation of cracks in self-assembled viscoelastic fluids, which are made of surfactant micellesreversibly linked by telechelic polymers. The morphology of the micelles can be continuously tuned, from spherical to rodlike towormlike, thus producing transient double networks when the micelles are sufficiently long and entangled and transient singlenetworks otherwise. For a single network, we show that cracks nucleate when the sample deformation rate involved is comparable tothe relaxation time scale of the network. For a double network, by contrast, significant rearrangements of the micelles occuras a crack nucleates and propagates. We show that birefringence develops at the crack tip over a finite length, ξ, whichcorresponds to the length scale over which micelle alignment occurs. We find that ξ is larger for slower cracks, suggesting anincrease of ductility.

An original technique that combines digital holography, dual illumination of the sample and cleaning algorithm 3D reconstruction is proposed. It uses a standard transmission microscopy setup coupled with a digital holography detection. The technique is 4D, since it allows to determine, at each time step, the 3D locations (x, y, z) of many moving objects that scatter the dual illumination beam. The technique has been validated by imaging the microcirculation of blood in a fish larvae sample (the moving objects are thus red blood cells RBCs). Videos showing in 4D the moving RBCs superimposed with the perfused blood vessels are obtained.

Morphological transformations of living cells, such as shape adaptation to external stimuli, blebbing, invagination, or tethering, result from an intricate interplay between the plasma membrane and its underlying cytoskeleton, where molecular motors generate forces. Cellular complexity defies a clear identification of the competing processes that lead to such a rich phenomenology. In a synthetic biology approach, designing a cell-like model assembled from a minimal set of purified building blocks would allow the control of all relevant parameters. We reconstruct actomyosin vesicles in which the coupling of the cytoskeleton to the membrane, the topology of the cytoskeletal network, and the contractile activity can all be precisely controlled and tuned. We demonstrate that tension generation of an encapsulated active actomyosin network suffices for global shape transformation of cell-sized lipid vesicles, which are reminiscent of morphological adaptations in living cells. The observed polymorphism of our cell-like model, such as blebbing, tether extrusion, or faceted shapes, can be qualitatively explained by the protein concentration dependencies and a force balance, taking into account the membrane tension, the density of anchoring points between the membrane and the actin network, and the forces exerted by molecular motors in the actin network. The identification of the physical mechanisms for shape transformations of active cytoskeletal vesicles sets a conceptual and quantitative benchmark for the further exploration of the adaptation mechanisms of cells.

We experimentally and theoretically study the variety of elastic deformations that appear when colloidal inclusions are embedded in thin wetting films of a nematic liquid crystal with hybrid anchoring conditions. In the thickest films, the elastic dipoles formed by particles and their accompanying defects share features with the patterns commonly observed in liquid crystal cells. When the film gets thinner than the particles size, however, the capillary effects strongly modify the appearance of the elastic dipoles and the birefringence patterns. The influence of the film thickness and particles sizes on the patterns has been explored. The main experimental features and the transitions observed at large scale—with respect to the inclusions’ size—are explained with a simple two-dimensional Ansatz, combining capillarity and nematic elasticity. In a second step, we discuss the origin of the variety of observed textures. Developing a three-dimensional Landau-de Gennes model at the scale of the particles, we show that the presence of free interfaces and the beads confinement yield metastable configurations that are quenched during the film spreading or the beads trapping at interfaces.

We form films of carboxylated polystyrene particles (C-PS) at the air water interface and investigate the effect of subphase pH on their structure and rheology by using a suite of complementary experimental techniques. Our results suggest that electrostatic interactions drive the stability and the structural order of the films. In particular, we show that by increasing the pH of the subphase from 9 up to 13, the films exhibit a gradual transition from solid to liquidlike, which is accompanied by a loss of the long-range order (that characterizes them at lower values of pH). Direct optical visualization of the layers, scanning electron microscopy, and surface pressure isotherms indicate that the particles deposited at the interface form three-dimensional structures involving clusters, with the latter being suppressed and a quasi-2D particle configuration eventually reached at the highest pH values. Evidently, the properties of colloidal films can be tailored significantly by altering the pH of the subphase.

We study the dynamics of individual polystyrene ellipsoids of different aspect ratios trapped at the air-water interface. Using particle tracking and in situ vertical scanning interferometry techniques we are able to measure translational drags and the protrusion in air of the ellipsoids. We report that translational drags on the ellipsoid are unexpectedly enhanced: despite the fact that a noticeable part of the ellipsoid is in air, drags are found larger than the bulk one in water.
