The development of new diagnostic technologies based on the light scattering and autofluorescence properties of den- tal tissues is required to improve the diagnostic ability of ini- tial caries lesions earlier than previously done and promot- ing the potential of treatment without surgical intervention. The aim of this study is to correlate fluorescence-based re- sults provided by multiphoton microscopy (MPM) with con- focal Raman microscopy records using phosphate level at 960 cm–1 and the organic matrix at ∼2,931 cm–1 in healthy and demineralized human enamel. Measurements on 14 teeth were made using two incident lights of different wave- lengths, released by confocal Raman microscopy and MPM. Raman phosphate peak intensity at 960 cm–1 along with or- ganic to mineral ratio at (2,931/430 cm–1) and nonlinear opti- cal signals (second harmonic generation [SHG] and intrinsic two-photon excited fluorescence [I2PEF]) were recorded from the demineralized and healthy enamel sites. Raman spectral maps showed that the higher the organic/mineral ratio in the demineralized enamel, the lower the intensity of mineral component in the same zone. MPM revealed new optical indicators of carious lesion as shown by the presence of a red-shifted fluorescence peak in the 650- to 750-nm area of the fluorescence spectrum of demineralized enamel. Moreover, on sample regions with insignificant autofluores- cence, the emergence of the SHG signal could be noted. By comparing I2PEF images with the structural motifs observed by the confocal Raman imaging system, the morphological similarity of the acquired images was quite evident. Any change in the I2PEF spectra reflects alterations in the chem- ical composition of enamel. These findings may provide an important basis for potentially valuable applications of pho- tonic tools in the clinical diagnosis of tooth pathological con- ditions, besides exposing the fundamental role of organic matrix in enamel integrity and reparation.

Nitrogen-vacancy centers in diamonds possess an electronic spin resonance that strongly depends on temperature, which makes them efficient temperature sensors with sensitivity down to a few mK/√Hz. However, the high thermal conductivity of the host diamond may strongly damp any temperature variations, leading to invasive measurements when probing local temperature distributions. In the view of determining possible and optimal con gurations for diamond-based wide- eld thermal imaging, here, we investigate both experimentally and numeri- cally the effect of the presence of diamonds on microscale temperature distributions. Three geometrical con gurations are studied: a bulk diamond substrate, a thin diamond layer bonded on quartz, and diamond nanoparticles dispersed on quartz. We show that the use of bulk diamond substrates for thermal imaging is highly invasive in the sense that it prevents any substantial temperature increase. Conversely, thin diamond layers partly solve this issue and could provide a possible alternative for microscale thermal imaging. Dispersions of diamond nanoparticles throughout the sample appear as the most relevant approach as they do not affect the temperature distribution, although NV centers in nanodiamonds yield lower temperature sensitivities than bulk diamonds.

In this course we will discuss the synthesis and the characterizations of nanomaterials ina liquid environment. Nanoparticles and nanostructured materials are indeed oftensynthesized in solutions via bottom-up routes. In these approaches, chemical and physicalforces operating at the nanoscale are used to assemble basic units into larger structures ofcontrolled morphology.In a first step we will review the most important colloidal interactions and the mechanismsleading to the broad diversity of self-organized structures found in complex fluids. In asecond step we will examine in detail a few examples where monodisperse nanoparticlesor well defined nanostructured solids are obtained via reduction of salts, sol-gelapproaches etc… and how they can be stabilized in solutions.Finally the last sequence of the course will be dedicated to the description of classic tools(such as light scattering techniques, spectroscopies …) that can be used to characterizenanomaterials and follow their formation in solutions. We will present some techniquesand their underlying mechanisms through a few basic examples where the size, thestructure or the properties of the nanomaterials are monitored during their synthesis.

Wheat gluten, one of the most complex viscoelastic protein networks in nature, is unique to get the specific texture of bread. Due to its complex protein composition, its insolubility in most solvents and the very high molar mass of half of the proteins (glutenin, the other half being gliadin), the architecture of the network is still not well understood. In this work, we have investigated model gluten protein extracts with contrasted compositions in glutenin and gliadin solubilized in a mild chaotropic solvent: ethanol/water (50/50 v/v). The samples display a liquid-liquid phase separation with an upper critical solution temperature that depends on the protein composition. The phase diagrams are consistent with the presence of supramolecular assemblies of proteins. To confirm the presence of these assemblies and fully characterize the objects dispersed in ethanol/water, we have used an asymmetrical flow field-flow fractionation (AsFlFFF) setup coupled with differential refractive index, multi-angle light scattering and dynamic light scattering detections to probe very dilute protein suspensions. We have identified three classes of objects, with distinctive molar mass, characteristic size and conformation: protein monomers, polymeric structures, and very loose protein assemblies with molar mass larger than 2.10 6 g/mol. A molecular characterization of the species by size exclusion chromatography in a denaturing solvent shows that polymers and assemblies are mainly composed of glutenin and ω-gliadin. The high content of ω-gliadin, devoid of cysteines, indicates the importance of non-covalent interactions involved in protein assemblies and might play a major role in gluten rheology

Background: Partial arterial pressure of carbon dioxide (CO2) modulates cerebral blood flow through a vasoreactivity mechanism. Near infrared spectroscopy (NIRS) can be used to record these changes in cerebral hemodynamics. However, no laterality comparison of the NIRS signal has been performed despite being a prerequisite for the use of such a method in a vasoreactivity monitoring context. We propose to investigate the NIRS signal laterality in response to a CO2-inhalation-based hypercapnia paradigm in healthy volunteers.Methods: Eleven healthy volunteers (6 women, 5 men, mean age: 31 ± 11) underwent a 3-block-design inhalation paradigm: normoxia (5min, "baseline") - hypercapnia (2min, "stimulation") - normoxia (5min, "post-stimulation"). NIRS signal was measured using a two-channel oximeter (INVOS 5100C, Medtronic, USA) with sensors placed symmetrically on both left and right sides on each subject's forehead. Additional heart rate (HR) monitoring was performed simultaneously. Based on the NIRS mean signal pattern, an a priori model of parametric identification was applied for each channel to quantify parameters of interest (amplitude, time delay, excitation and post-stimulation time) for each inhalation block.Results: HR increased significantly during the stimulation block. The quality of the model was satisfactory: mean absolute errors between modeled and experimental signals were lower than the resolution of the device. No significant lateralization was found between left and right values of most of the parameters.Conclusion: Due to the lack of lateralization, this parametric identification of NIRS responses to hypercapnia could bring light to a potential asymmetry and be used as a biomarker in patients with cerebrovascular diseases.

Background; Muscle weakness following critical illness is the consequence of loss of muscle mass and alteration of muscle quality. It is associated with long-term disability. Ultrasonography is a reliable tool to quantify muscle mass, but studies that evaluate muscle quality at the critically ill bedside are lacking. Shear wave ultrasound elastography (SWE) provides spatial representation of soft tissue stiffness and measures of muscle quality. The reliability and reproducibility of SWE in critically ill patients has never been evaluated.Methods; Two operators tested in healthy controls and in critically ill patients the intra- and inter-operator reliability of the SWE using transversal and longitudinal views of the diaphragm and limb muscles. Reliability was calculated using the intra-class correlation coefficient and a bootstrap sampling method assessed their consistency.Results: We collected 560 images. Longitudinal views of the diaphragm (ICC 0.83 [0.50–0.94]), the biceps brachii (ICC 0.88 [0.67–0.96]) and the rectus femoris (ICC 0.76 [0.34–0.91]) were the most reliable views in a training set of healthy controls. Intra-class correlation coefficient for inter-operator reproducibility and intra-operator reliability was above 0.9 for all muscles in a validation set of healthy controls. In critically ill patients, inter-operator reproducibility and intra-operator 1 and 2 reliability ICCs were respectively 0.92 [0.71–0.98], 0.93 [0.82–0.98] and 0.92 [0.81–0.98] for the diaphragm; 0.96 [0.86–0.99], 0.98 [0.94–0.99] and 0.99 [0.96–1] for the biceps brachii and 0.91 [0.51–0.98], 0.97 [0.93–0.99] and 0.99 [0.97–1] for the rectus femoris. The probability to reach intra-class correlation coefficient greater than 0.8 in a 10,000 bootstrap sampling for inter-operator reproducibility was respectively 81%, 84% and 78% for the diaphragm, the biceps brachii and the rectus femoris respectively.Conclusions: SWE is a reliable technique to evaluate limb muscles and the diaphragm in both healthy controls and in critically ill patients.

We reconsider our former determination of the chiral quark condensate $\langle \bar q q \rangle$ from the related QCD spectral density of the Euclidean Dirac operator, using our Renormalization Group Optimized Perturbation (RGOPT) approach. Thanks to the recently available {\em complete} five-loop QCD RG coefficients, and some other related four-loop results, we can extend our calculations exactly to $N^4LO$ (five-loops) RGOPT, and partially to $N^5LO$ (six-loops), the latter within a well-defined approximation accounting for all six-loop contents exactly predictable from five-loops RG properties. The RGOPT results overall show a very good stability and convergence, giving primarily the RG invariant condensate, $\langle \bar q q\rangle^{1/3}_{RGI}(n_f=0) = -(0.840_{-0.016}^{+0.020}) \bar\Lambda_0 $, $\langle\bar q q\rangle^{1/3}_{RGI}(n_f=2) = -(0.781_{-0.009}^{+0.019}) \bar\Lambda_2 $, $\langle\bar q q\rangle^{1/3}_{RGI}(n_f=3) = -(0.751_{-.010}^{+0.019}) \bar\Lambda_3 $, where $\bar\Lambda_{n_f}$ is the basic QCD scale in the \overline{MS} scheme for $n_f$ quark flavors, and the range spanned is our rather conservative estimated theoretical error. This leads {\it e.g.} to $ \langle\bar q q\rangle^{1/3}_{n_f=3}(2\, {\rm GeV}) = -(273^{+7}_{-4}\pm 13)$ MeV, using the latest $\bar\Lambda_3$ values giving the second uncertainties. We compare our results with some other recent determinations. As a by-product of our analysis we also provide complete five-loop and partial six-loop expressions of the perturbative QCD spectral density, that may be useful for other purposes.
